One-shot roles can only utilize their special abilities once during the course of the game. The most common use of this role is to limit the power of Vigilantes, but there can also be one-shot Cops, one-shot Bulletproofs, one-shot Governors, etc. Most roles can be adapted to be "one-shot" in nature.

In many cases, when a roleblocker targets a person using a one-shot power, the power is considered used and gone.
